2x 650 is maximum you can get away with.
With 650 flows, estimate is that mean read length is 346, and 90% of reads should make it to 325.

Ion Torrent Metagenomics flows
Hello all, I've been looking for optimalization of my metagenomics sequencing process.
I usually use 316chip on PGM and as default in IonTorrent 16S metagenomics workflow inumber of flows is setted to 850. It results with half of volume of every dNTP left in bottles and I'm afraid of runnning next sequencing without replacement them to new. It is really increasing my sequencing cost because I need to use new inicialization chemistry for each sequencing. I use about 400 bp length fragments but in real its average is about 250 bp(as I can see in the end sequencing summary). Can I decrease number of flows to maybe about 650 to reach two sequencing processes with one inicialization?Tags: None
