I am trying to establish a protocol to generate 16S libraries using the Ion Amplicon Library Preparation (Fusion Method) that does not rely on the expensive 16S metagenomics + fragment library plus kits from life tech.
Just wondering whether it would be possible to generate indexed 16S libraries by a single 1 step PCR by just adding the sequence of the Ion Xpress barcode (or others if the case) on each primer pair. This way, I would have 2 fused primer pairs (P-16S FW, P-16S REV, Barcode-16S FW, Barcode-16S REV) for each sample.
Any help on this would be much appreciated!
