    Hi Ben3 thank you for your help. I have a question about Lib prep kits.

    llumina® DNA PCR-Free Prep, Tagmentation (24 Samples)...this kit contains everything for preparing 24 sample together Right? But if I want prepare 12 sample can I split this kit?

    Multiplexing Up to 384 unique dual (UD) combinations and 96 comb[/TD]: what does mean?

    Yeah if you wanted to only prep a couple samples out of a 96 kit, then you could do that. I've done it plenty of times. I'm not sire what you mean about the probe for enrichment. Can you give me more details about the probe?

    And a UDI is the index sequence. It's the barcode that goes on your DNA sequence so you can keep track of the samples. The UMI is another barcode. It helps keep track of individual molecules WITHIN a sample. So it is used a lot of times for checking mutations or filtering out PCR duplicates. Whenever you make libraries, you usually have to amplify them and some of the pieces will get sequenced multiple times. The UMI is used to filter out the duplicates.

    Ok Thank you really so much. So I have another 2-3 dubts. Ahahaha.... all lib prep kit Illumina are divisibile so if i have kit for 94 sample and i want to prepare 6 sample can i?is it the same also for probe for enrichment? And what is the difference between UDI and UMI. Thank you really much.

    You are correct with the sample kit. That should have everything you need for 24 samples and you can save the rest for another 12 samples. Just make sure you have all the components because Illumina ships kits in multiple boxes.

    Also, multiplexing just means adding more samples together. This is saying that the type of adapters it uses can allow for up to 384 samples to be run on the sequencer at the same time. Each adapter is like a barcode. It helps you identify individual samples during data analysis. Without them, all of the sequence data (reads) would be mixed together.
