  • Roche 454 Amplicon Variant Analysis Help

    I need a bit of help with my attempts at using Amplicon Variant Analysis (AVA).

    I have a consistent problem when attempting a new analysis. I have correct references and annotation files imported. I have good data files (sff). When I attempt to build the associations everything appears correct when I compare what I set up to a previous successful analysis, but when I attempt to compute it, it says there is no active data to trim.

    It acts as if there is no data in the associations, but it is obvious I have active sff files associated. When others in my lab attempt the same analysis using my same imported files and data, it will work. I try to copy it exactly, but it doesn't work for me.

    In addition, I've tried to add new data to previous successful analysis files, but the software doesn't recognize the additional data and will only ever rerun the previous analysis.

    There must be some minutia that I'm missing while building the associations and attempting the analysis. Any idea what my problem is? I can't even begin to guess what's up, I'm not missing any major steps. Is it really that sensitive to the order I do things?

    Did you check a box "Active". In project click "Read Data" tab, you will find menus "name" "annotation" "group" and "active". Check the box and try it.
