I have a question about the reaction primers of ABI Solid sequencing.
As all you know that Solid use ligation-based sequencing. From different places on the web, two different sets of primers were illustrated.
One set is primers like: nnnXXzzz
The other set contains primers like: XXnnnzzz
XXs are the dinucleotide that gives the color space read.
zzzs are the nucleotides that are trimmed off before next ligation.
Anyone knows which one of the above is currently used?
