    Help.. RNA-seq validation options.

    Hello all!

    I would like to know if someone could guide me with the following: I am finalizing a transcriptomic analysis, however, due to the current health contingency I have not been able to access the lab to validate the experiment by qPCR. Do any of you know of another way to validate this type of study or do you know of any journal that accepts this work without the need for qPCR validation? This situation for COVID-19 is getting longer and we do not see the moment when we can return to the lab, so we are looking for options to validate it or to publish without the need of using qPCR. I appreciate your responses in advance.

    Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.

    Data variability is still an issue in sequencing technologies despite the advances in reproducibility and accuracy of these platforms. But the problem does not originate in the sequencing itself, but in the previous steps, before the sample reaches the sequencer.


    The first step is collection, followed by preservation and sample preparation for analysis. Most scientists overlook those steps, but not being careful might just be skewing the experiment’s results.
