I start my 1st automated experiment with TruSeq stranded total RNA. Unfortunately, my pipetting platform doesn't have a cooling block (I didn't choose this option when I bought it), my question is :
- For the "Synthesize 1st strand cDNA" and "Adenylate 3' Ends" steps, basically, we need to incubate samples at 70°C then hold at 4°C. Can I process directly the next step without this 4°C, or use a cooling block passive (not controled temperature, so the temperature can be 4, 6, 10°C...)?
Does anyone faced this problem? Help me please.
