The aim of my project is to do a metagenomic analysis on microbial communities (bacteria and DNA/RNA viruses). I'm very much a newbie and I need some advice on nucleic acid extractions/sample prep as well as choosing an appropriate platform.
I've come across column based kits that can be used to extract RNA and DNA simultaneously. I plan on using this followed by whole genome and transcriptome amplification kits to have sufficient starting material. Does this sound reasonable?
Any suggestions on the sequencing depth and coverage, suitable to detect less abundant bugs?
