    A question about install RSEQtools

    Hi everyone,

    I am interesting in RSEQtools,but I have some problem while install.

    In the begin,I have already installed gsl,and I download source code RSEQtools-0.6.tar.gz

    After I set environment variables,I cd to bios and keyin make,but show error as following:

    Makefile:6: /usr/local/bin//biosdefs.make: No such file or directory
    make: *** No rule to make target `/usr/local/bin//biosdefs.make'. Stop.

    My problem is I can't find biosdefs.make in my gsl,I think gsl 's version I installed is different as official.

    But I don't which version is official usage?

    Thanks!!!!!!

    Best Regard!

    I am unable to use sam2mrf provided by prebuilt binaries of RSEQtools, can anybody pls help?
    The command I tried was "sort -r file.sam | sam2mrf > file.mrf"
    as specified in the manual for the alignment file generated by tophat but it didnt work.

    please reply soon.

      Hi,

      I had the same problem until I realized that I had to export the path variables from my bash.

      Previous .BASHRC settings:
      #RSEQtools
      BIOINFOCONFDIR=~/Programs/RSEQtools/bios/conf
      BIOINFOGSLDIR=~/Programs/gsl-2.4

      Changed to:
      #RSEQtools
      export BIOINFOCONFDIR=~/Programs/RSEQtools/bios/conf
      export BIOINFOGSLDIR=~/Programs/gsl-2.4

      [I]Settings were saved[\I]

      source ~/.bashrc
      make

        Nine Things a Sample Prep Scientist Thinks About Before Sequencing
        by SEQadmin2

        I’m not a sequencing expert. I’m a purification scientist who uses NGS to evaluate workflows my group develops. With this perspective, we think about the sample first and the NGS workflow second. The sequencer is an exceptionally honest reporter, but it can only report on what you give it, so whether you get clean, interpretable data from an NGS workflow is largely determined before you begin.

        Here are nine questions we think about, in roughly the order they matter, before...

        From Collection to Sequencing: Why Sample Preparation and Preservation Define Sequencing Data
        by SEQadmin2

        Data variability is still an issue in sequencing technologies despite the advances in reproducibility and accuracy of these platforms. But the problem does not originate in the sequencing itself, but in the previous steps, before the sample reaches the sequencer.

        The first step is collection, followed by preservation and sample preparation for analysis. Most scientists overlook those steps, but not being careful might just be skewing the experiment’s results.
