Reconstructing a full genome from 1000 genome project

    Hi there,

I am trying to reconstruct whole DNA sequence of an individual (HG00096) from 1000 genome project and save it in a fasta file. I have examined ftp.1000genomes.ebi.ac.uk and found unmapped bam files under directory ftp.1000genomes.ebi.ac.uk/vol1/ftp/data/HG00096/alignment for individual HG00096. Although i searched forum, I kinda got stuck at this point.

1- Is this the right file containing different parts of individual HG00096 from reference?
2- If so, where can i find the suitable reference genome to work with this bam file?
3- With which tool can i combine reference genome and unmapped parts of individual HG00096?

Sorry for the lousy question but i am kinda new in this job.

    Thanks in advance.

All the raw FASTQS that generated the bam file ( the aligned file) should be there too somewhere. It might be helpful if you explained why you are trying to do that.
I've found it a bit of a minefield, but here are directions on the 1000 genome bite to hopefully point you in the right direction. Failing that, they have a help email address.

New Genomics Technologies Take Aim at Long-Standing Limits
      by SEQadmin2

Researchers using sequencing and genomics tools often have to make trade-offs. They can choose between speed or scale, short reads or long-range information, or targeted panels or a view of the whole transcriptome. New technologies that have been released this year are built to address those tough choices.

We asked six companies the same four questions to learn about their latest products. The new technologies bring a lot to the table, including rethinking sequencing

How Immunogenomics Decodes Immunity’s Genetic Blueprint
      by SEQadmin2

The immune system’s power comes from its genetic diversity, allowing myriad threats to be neutralized through first recognizing foreign antigens. That diversity is also what makes the immune system so difficult to study. Recent advances in sequencing technology and computational biology, however, are giving researchers new tools to understand immune responses and immune-related diseases in greater detail.

This convergence of genetics, immunology, and computation...
