Have you added boost to your path? Maybe you should try the following:

    export LD_LIBRARY_PATH=/boostdir/boost/dist/lib/

      Thanks, iris_aurelia

      you mean..
      /boostdir/boost/dist/lib/
      is the boost prefix directory??

      I did like this:
      export LD_LIBRARY_PATH=/usr/local/bin/lib/

      but still error=127 is comming.

and i dont know what the "LD_LIBRARY" is.

Yes, you should give it the path where your boost files are stored.

I just want to check install stat, so I using test file.

            following is the tophat command:
            tophat -r 20 test_ref reads_1.fq reads_2.fq

than i got the error message (error=127).

And my boost prefix directory(/usr/local/bin/lib/) have

            libboost_chrono.a libboost_locale.a libboost_prg_exec_monitor.a libboost_system.a libboost_context.a libboost_math_c99.a libboost_program_options.a libboost_test_exec_monitor.a
            libboost_date_time.a libboost_math_c99f.a libboost_python.a libboost_thread.a libboost_exception.a libboost_math_c99l.a libboost_random.a libboost_timer.a libboost_filesystem.a libboost_math_tr1.a libboost_regex.a libboost_unit_test_framework.a
            libboost_graph.a libboost_math_tr1f.a libboost_serialization.a
            libboost_wave.a libboost_iostreams.a libboost_math_tr1l.a libboost_signals.a libboost_wserialization.a

these kind of files, do u know this is right? I dont know whether it correct or not...

            thank you guys!

It seems you don't have libboost_thread.so.1.52.0 in your directory.
If you are not familiar with linux I suggest you install a pre-compiled binary release, instead of build it from source. So you don't need boost lib anymore.
              this is the tutorial:

From Collection to Sequencing: Why Sample Preparation and Preservation Define Sequencing Data
                by SEQadmin2

Data variability is still an issue in sequencing technologies despite the advances in reproducibility and accuracy of these platforms. But the problem does not originate in the sequencing itself, but in the previous steps, before the sample reaches the sequencer.

The first step is collection, followed by preservation and sample preparation for analysis. Most scientists overlook those steps, but not being careful might just be skewing the experiment’s results.

                by SEQadmin2

With the launch of new single-cell sequencing platforms in 2026, the field stands at an exciting inflection point. This article surveys the most impactful advances in the field and discusses how they’re reshaping research in cancer, immunology, and beyond.

                Introduction

Single-cell sequencing technologies have undergone remarkable advances over the past decade, transitioning from low-throughput experimental approaches to highly scalable platforms capable of...

Environmental Genomics in the Age of NGS: From Microbes to Conservation Strategies
                by SEQadmin2

Studying ecosystems means dealing with complex, multi-species communities that are hard to observe at scale. This complexity, however, hides many important questions to be answered, from how biogeochemical cycles work and how climate change can affect species distribution to how conservation strategies can work best.

Genomics, particularly since the expansion of NGS, has transformed ecosystem ecology. By sequencing environmental DNA, we can now assess biodiversity without direct...
