    Installing Mira 4.

    Hi,

ok I'm feeling very stupid, I have literally spent a whole day tryin gto install Mira 4 with no luck.

I installed Boost libraries with the fisrt instructions i found (none on boost website helpfully):

but that put them in the wrong directory for the mira compiler.. so
    ./configure --with-boost-libdir=/home/susan/usr/lib

does not work.. always get an error:
configure: error: Stopping here as BOOST thread lib must be available at this stage. Sorry.

Does anyone PLEASE know how to install Mira...???? seems that a program that can't be installed easily is pretty useless..!!

    Thanks,

    S.

As MIRA4 install pages says it will only work with fully installed BOOST libraries so try

    Code:
    $ sudo apt-get install libboost-all-dev
Have you considered getting the pre-compiled binaries and using them? http://sourceforge.net/projects/mira...s/MIRA/stable/

Hi, erm.. can you point me to where they are? I can only see Mira 4 here:

The manual says there is a bin folder, but there isn't one in there. I'll try and download it again.

      thanks.

Ok, I got another error, saying 'Flex' wasn't installed..

        Then got:
"Ooops, we might encounter some problems in compilation/linking. Please give
it a try and report success or failure."

Then, predictably, make fails with a lot of errors.

Is there a prize for getting this installed?

        S.

          Pre-compiled binaries are here: http://sourceforge.net/projects/mira...s/MIRA/stable/

Sorry about not linking the correct files before.

Reply to Nine Things a Sample Prep Scientist Thinks About Before Sequencing
            by GATTACAT
Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.

Nine Things a Sample Prep Scientist Thinks About Before Sequencing
            by SEQadmin2

I’m not a sequencing expert. I’m a purification scientist who uses NGS to evaluate workflows my group develops. With this perspective, we think about the sample first and the NGS workflow second. The sequencer is an exceptionally honest reporter, but it can only report on what you give it, so whether you get clean, interpretable data from an NGS workflow is largely determined before you begin.

Here are nine questions we think about, in roughly the order they matter, before...

From Collection to Sequencing: Why Sample Preparation and Preservation Define Sequencing Data
            by SEQadmin2

Data variability is still an issue in sequencing technologies despite the advances in reproducibility and accuracy of these platforms. But the problem does not originate in the sequencing itself, but in the previous steps, before the sample reaches the sequencer.

The first step is collection, followed by preservation and sample preparation for analysis. Most scientists overlook those steps, but not being careful might just be skewing the experiment’s results.
