  • Opposite directions in homologous proteins

I am analyzing protein uncharacterized bacteria so far, which have a transmembrane domain and a coiled coil domio with low identity that other characterized protein of another bacteria phylum. In prediction programs, I observed that this protein has a Coiled coil domain in C' terminus, right in the periplasm, contrary to this, the protein that is apparently homologous has the coiled-coil in the N' terminus also targeted to the periplasm.
I can not find examples of homologous proteins which have opposite directions.

How sure are you that these are actual proteins?

Because both coils and transmembrane doamins can come as well from wrong annotations.
Especially some XXX in the genome sequence will lead to coils, and even intergenic regions will display some sort of transmembrane or periplasmic domains just be chance.

      Hello Dear
Yes, I am sure that this proteins are well annotated.
I attached an scheme of protein topology.

My next guess would be that in both cases either start or end are missing.

        If not...good luck, seems interesting.

Recent Advances in Sequencing Analysis Tools
          by seqadmin

The sequencing world is rapidly changing due to declining costs, enhanced accuracies, and the advent of newer, cutting-edge instruments. Equally important to these developments are improvements in sequencing analysis, a process that converts vast amounts of raw data into a comprehensible and meaningful form. This complex task requires expertise and the right analysis tools. In this article, we highlight the progress and innovation in sequencing analysis by reviewing several of the...

Essential Discoveries and Tools in Epitranscriptomics
          by seqadmin

The field of epigenetics has traditionally concentrated more on DNA and how changes like methylation and phosphorylation of histones impact gene expression and regulation. However, our increased understanding of RNA modifications and their importance in cellular processes has led to a rise in epitranscriptomics research. “Epitranscriptomics brings together the concepts of epigenetics and gene expression,” explained Adrien Leger, PhD, Principal Research Scientist...
