    DNA extraction on the EasyMag

    Hi everyone,

    We have been doing our DNA extraction on the EasyMag by Biomerieux from stool, breast milk, skin, oral, and rectal swabs. Our samples are preserved in Stratec Stool DNA Stabilizing Solution (also part of PSP Spin Tool DNA Plus Kit). However, we've discovered recently that this buffer seems to react with the EasyMag lysis buffer, forming viscous white layer, that may be contributing with the failed runs we've been having with the instrument.

    My questions:
    1) Have any of you had this experience with the EasyMag?
    2) If so, how did you resolve the problem? Offboard lysis would allow us to vortex the preservation buffer+lysis buffer, which does improve the consistency of the sample, but it is still cloudy and I wonder if it will still impair the machine in the long run by causing a buildup in the lines...
    3) Is there another good DNA preservation buffer (especially for stool samples) that you have used that is more compatible with the EasyMag?
    4) The Stratec Stool DNA Stabilizing solution also has lysis ability; I wonder if using this buffer alone without using the EasyMag lysis buffer would still result in a successful extraction on the machine?
