    Estimating Sequencing Coverage with MiSeq

    Hi everyone,

    I am new to sequencing and am wondering if it would be possible to perform de novo assembly using MiSeq, for a 35 MB fungal genome ?
    Technically speaking, how could we increase the coverage with the Nextera kit, already purchased for this purpose ?
    Is it feasible to run the same sample many times to increase the fastq file, I mean the ouput ?
    Sorry, if I am saying stupid things but this is so important to me,

    Once you make the library you can run it as many times as you want to get additional data. You would still need to buy sequencing kits or pay for the runs (as the case may be).
