    Ultimately it is arbitrary. I think the E.coli genome project used a 0 near the replication origin, but that need not be followed.

    Another possible landmark would be a well known restriction site, perhaps chosen from some previous work on the organism such as a physical map.

    most of bacteria genome are circle, but in database most are saved in line. I mean they have first position and last position. I used to believe the ori position, which is replication start point, was defined as first position, now i find i am wrong. how is this defined. any help will appreciated.
