    Barcode of plants

    Dear All,

    part of my work will be a study on the cpDNA for barcoding of plants, I need a variable region within conserved primers. I think I might look for a conserved gene who presents (in the nucleotide sequence) an intron in within the active site of the protein... does anyone know a chloroplast gene who responds to my requests?

    cpDNA extraction

    dear all, I have one more question, I do not have in the labs here the instruments and the expertize to carry a cpDNA extraction (it seems that an ultracentrifuge is needes according to the protocol of jansens et al 2005). So I should send my samples to someone who extract cpDNA as a routine work. Could you give me any advice?

      Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
