    Tissue Preservation

    Hi,
    I have access to samples that have been preserved in 70% EtOH for 4 years. Unsure of the temperature. These present a historic population that no longer exists, so are high value. Will it be possible to get data using GBS or RAD from these samples? FYI I have access to a reference genome.

    I think the first step would be to extract DNA and check the quality. If quality is low then a DNA repair enzyme mix can be used. GBS then may be should be done on individual samples and equimolar pooled for sequencing after PCR to obtain similar number of reads for samples.

      Thanks. I have requested the material in order to see what quality the DNA is in. Thanks for the advice on running the samples individually.
