    Goal to optimize library prep for HiSeq

    My goal is to find a way to make library prep faster and cheaper. Any ideas are welcome!

    A few random thoughts

    1) The Long Fragment Read protocol published recently from Complete Genomics was interesting, and if their accounting is correct was quite inexpensive

    2) Transposon generation of libraries ("tagmentation") has proven valuable & has attracted a number of variations (e.g. using it for mate-pairs, combining it with digestion & ligation for expanded barcoding). What else can be done with this?

    3) Is there a methodology outside the transposon space (especially the patent space!) which has similar characteristics -- one tube, low input material, combining multiple steps?

    4) What can be done with either droplet or molded microfluidics? Relatively underexplored area -- though one version has been commercialized as Mondrian.

      "homebrew" for library prep

      I'm mostly interested in making a "homebrew" of enzymes to perform end-repair, A-tail, and adapter ligation. I would like to be able to avoid cleanups between each reaction too. I know Epicentre has an end-repair/ligation kit that omits having to do a cleanup in between if it is a blunt ligation. I also know NEB has some good enzymes that I could start working with. Do you know any other good enzyme companies or kits that would help me with this?

        I'm not in the lab, but two companies that seem to offer interesting enzymes are Kapa and Lucigen.
