    hello and DNASE question!

    hi everybody!

    this is my first post in this forum... i'm a phd student, i work either in France and in Italy! i'm going to work on RNA sequencing to analyze differentially genes expression in vivo and in vitro in Gram Positive bacteria.

    I have a question:i gonna do a Dnase treatment after total RNA extraction using TURBO DNASE (invitrogen) ; do you think i have to re-extract RNA with phenol-cloroform after the DNASE treatment to inactivate DNASE or it's enough to inactivate it with high temperature and clean the RNA with the RNA CLEAN AND CONCENTRATOR kit from Zymoresearch?

    It depends how clean or how good you want your RNA to be. I would recommend the RNA cleanup kit. I usually do ethanol precipitation after DNase digestion because I need to concentrate it anyway.

      actually i don't exactly how the RBA has to be...it's the first time i do the RNA SEQ, i suppose it has to be clean and good quality as much as i can !
