    Hello world !

    Hello there !

This seems like a great forum, I can't believe I didn't found ot about it earlier.

My name is Sébastien, I am a microbiologist currently doing quite a bit of metagenomics. I started using NGS 2-3 years ago, with 454 16S sequencing of bacterial communities. I am still working on microbiota (human and animal models related), but moving on Illumina/Ion Torrent 16S and shotgun (metagenomics).

I have a bit of experience in QIIME, bio statistics (R, PCOrd...) and microbiology in general. I'll be happy to share my experience, so don't hesitate to ask !

See you soon on the forum !

    Hello Sebastien

    I hope you are well.
Would you be able to advise me on the best way to proceed with analysis
of my 454 data. I am carrying out research on the microbiota of chicken GI tract.
I am having some difficulties in developing a pipeline for data analysis.
I have virtually no experience in Bioinformatics.

    Cheers

    Colum

Nine Things a Sample Prep Scientist Thinks About Before Sequencing
      by SEQadmin2

I’m not a sequencing expert. I’m a purification scientist who uses NGS to evaluate workflows my group develops. With this perspective, we think about the sample first and the NGS workflow second. The sequencer is an exceptionally honest reporter, but it can only report on what you give it, so whether you get clean, interpretable data from an NGS workflow is largely determined before you begin.

Here are nine questions we think about, in roughly the order they matter, before...

From Collection to Sequencing: Why Sample Preparation and Preservation Define Sequencing Data
      by SEQadmin2

Data variability is still an issue in sequencing technologies despite the advances in reproducibility and accuracy of these platforms. But the problem does not originate in the sequencing itself, but in the previous steps, before the sample reaches the sequencer.

The first step is collection, followed by preservation and sample preparation for analysis. Most scientists overlook those steps, but not being careful might just be skewing the experiment’s results.
