  • cDNA Synthesis for AmpliSeq

    Hello Dear Community, I have 2 questions, I am working on AmpliSeq Myeloid Panel,
    1. Which cDNA Synthesis should I use with this kit?
    Illumina cDNA Synthesis or anyother vendor? If anyone have used?
    2. Which Tris-Hcl 7.0ph and 200mm should be purchased? if anyone can share a link with personal expierence?

    Hello there,

    It's been a while since I've run any AmpliSeq panels, but I do remember parts of the work. I think in general, we used the SuperScript IV First-Strand Synthesis System (Invitrogen). I'm pretty sure some other groups used the High-Capacity cDNA Reverse Transcription Kit and I've done other targeted sequencing projects with the RevertAid First Strand cDNA Synthesis Kit. That worked pretty well. But doesn't the panel have a recommendation in the protocol?

    Also, I know that we used to buy this tris from Sigma years ago. I'm sure you can still do the same and it will be fine.
