  • Average fragment length post-sonication

    Hi all,

    I have a question!
    Anyone knows what is the average fragment length obtained post-sonication when you are preparing a fragment library (from genomic DNA).

    For example, using GS Junior (Roche), I know that the range obtained post-shearing DNA (+ end repair and adaptors ligation) is: 600-900 bp. If I have this, I can continue working on library.

    So, what's the range for fragment library using Ion Torrent?

    Hi Soleulloa,

    I think that the answer is depending on the length that you will sequence.
    If you want to sequence 100, 200 or 300 bp you need to have a mean respectively to 120, 220 and 320 before adding adaptors.

    I'm not sure that with a range of 600 to 900 bp you will have success on ePCR with the Ion kit.

      Hi soleulloa,

      I've attached an image of the current library table. which highlights the target insert size for each sequencing read length. Parameters for the 400 base read methods will be added shortly.

      In addition, there's no need to rely on Google Translate to get these details All of the PGM User Guides are readily available on the Ion Community for all to access. In particular, the details you requested are on pp. 13 of the Ion Xpress™ Plus gDNA Fragment Library Preparation User Guide.
