I wonder if anyone have update on Sequel performance and how it compares to RSII.

There was a PacBio user group meeting in Baltimore (in last two weeks) where an update was given about Sequel performance (based on a note I saw from a FAS). Apparently that information has not been published but if someone on SeqAnswers was in attendance then they may be able to comment.

A few service providers had told me they expect to start providing Sequel services in June. I expect we should see some data soon?
At the UGM pacbio did mention they expect to start shipping their new chip at the end of the month along with a new loading protocol. So maybe service providers are waiting for that? I'm sure Sequel owners here would have a better idea of whats going on though.

Hi Brian,
is this with your previous or the new employer?
Thanks!

It is hard to tell but the description for this dataset seems to include verbiage about Sequel (not sure why the submitters would include that if this was a RSII): http://www.ebi.ac.uk/ena/data/view/SRX1684788
Most likely a red herring but who knows. Raw bax/bas.h5 files are available in case someone is curious (http://trace.ncbi.nlm.nih.gov/Traces...run=SRR3342814 under Download tab).Last edited by GenoMax; 07-22-2016, 05:21 AM.

We are facing some issues with our new sequal system installed recently. Pacbio team is unable suggest loading clear loading concentration of different sized library.
The out put varies with each run and the maximum we got till now is 3.2gb for 2 kb library.
For the higher sized library the runs will yield 0.2-0.9gb per SMRT.We are unable to find out where the problem lies. Also after completing each run the volume of libraries pickedup from the sample well is not consistent.
Pac bio suggestion for the loading conc is 50-60pM for any size library.
There is another suggestion from PacBio is that system and smrtlink need to be restarted every 24 days, Is it true?
Please help
