    ChIP-seq library complexity

    Hi all.
    I am new to the ChIP-seq and trying to ChIP genome-wide H3K27me3. Is there any methods or strategies for increasing the library complexity? Maybe shearing more using sonicator is one, but I am worrying about more sonication will hurt the K27 methylation and lead to poor IP. Or should I use more starting materials? Any thoughts would be grateful!

    Make sure you don't overamplify your library. We always aim for a final library concentration of 10 to 20 ng/ul. If your yield is higher than that you are overamplifying.
