    ATAC-Seq method starting with extracted DNA instead of cells

    I am new to try ATAC-seq method in my lab. The protocol from stanford has mentioned to start with 50,000 cells. I have already extracted DNA in my hand. is it possibly to use the gDNA for transpose instead of using cells? If yes, then what protocol should I follow? the same for cells? It will be great if anyone can suggest me about this. Thank you in advance,

    Surely the entire point of ATAC-Seq is to start with material that is still associated with nucleosomes so that the tagmentation protocol tells you something about chromatin accessibility. If you've done a gDNA prep and removed all traces of associated protein - exactly what do you expect to see?
