ChIP-seq with very small amount of tissue

    Hi,

We are trying to do ChIP with very small amount of tissue, like <10mg, which is tightly limited by the technical difficulty in collecting the tissue. Does anyone has experience doing ChIP with this little tissue, and obtained enough DNA for sequencing? If you would like to share some of your experience, or give me some advice, I would really appreciate it!! Thanks a lot!!

    Did you already read this?

    Nature Genetics 38, 835 - 841 (2006)
Epigenetic characterization of the early embryo with a chromatin immunoprecipitation protocol applicable to small cell populations

    and

I guess that after ChIP you will have to do a serious amplification of your IP material using WGA e.g.

      Hi,

Thanks for the info. Sorry maybe I did not make it clear, that the samples I deal with are actually pieces of tissues from microdissection and I had to freeze it down immediately after the dissection.

The paper linked above was using freshly isolated tissue/cells. It's really different. I am just wondering what would be the best way to homogenize, how much tissue to start with and how much chromatin you can get from it.

      Thanks,

Sorry. I have no experience with ChIP from tissues at all.

maybe the following link can help you:

you also could check out ChIP kits offered by various companies (Diagenode, Millipore etc)

Multiomic Liquid Biopsy: What DNA/RNA Panels Reveal That DNA Alone Can’t
          by SEQadmin2

Tumors contain complex mixtures of cell populations that evolve in response to treatment and host-specific factors. Tissue biopsies provide only static snapshots of a tumor’s state. But what if we could follow that evolution over time using a minimally invasive technique?

Liquid biopsies can detect tumor-derived material circulating in the bloodstream, providing a way to study treatment response, detect emerging resistance, and discover new biomarkers. Circulating

New Genomics Technologies Take Aim at Long-Standing Limits
          by SEQadmin2

Researchers using sequencing and genomics tools often have to make trade-offs. They can choose between speed or scale, short reads or long-range information, or targeted panels or a view of the whole transcriptome. New technologies that have been released this year are built to address those tough choices.

We asked six companies the same four questions to learn about their latest products. The new technologies bring a lot to the table, including rethinking sequencing
