  • Non-linear model for RNA-Seq (reads and expression levels)

    Hi there!

I’ve read the paper by J. Bullard (Sandrine Dudoit’s group) about expression level and its relation with the read counts in RNA-Seq:

Evaluation of statistical methods for normalization and differential expression in mRNA-Seq experiments

The equation (1) in the paper provides the relation between expected read counts and expression levels. This model is linear and is a good representation of the relation. However, I was wondering if there is any other paper(s) that studied a non-linear model, especially exponential relations.

    Any hint is appreciated!

    Hello,

I have recently read a paper
RNA-seq may replace gene expression microarrays in the near future. Using RNA-seq, the expression of a gene can be estimated using the total number of sequence reads mapped to that gene, known as the total read count (TReC). Traditional expression quantitative trait locus (eQTL) mapping methods, suc …

which utilized Poisson regression to analyze count data.
