SEQanswers June Challenge Has Begun!

  • microRNA study design - number of replicates?

    We are planning our first microRNA sequencing project using the Ion Torrent PGM. We want to determine changes in the microRNA profile after cell line treatment.

    First question would be, what would in your experience be the needed number of biological and technical replicates? Probably biological replicates are indispensable for an accurate study, but we are unsure whether technical replicates are also needed in an NGS setting?

    I think it's better to have more biological replicates than technical replicates. Sequencing runs are expensive so go for more biological replicates !
