DEXSeq fitDispersionFunction error (no replicates)

Hi,

I am trying to use DEXSeq for differential exons for my RNA-Seq data. We do not have any replicates in our dataset.

I tried fData(ecs)$dispersion <- .1 as mentioned at https://stat.ethz.ch/pipermail/bioco...er/042717.html

But, I still got the error Error in fitDispersionFunction(exset) :Failed to fit the dispersion function

Could you please let me know if you have suggestions for the same?

I would suspect that if you're going to set the dispersion manually then you shouldn't then try to reset it with fitDispersionFunction(exset).

I'll not bother commenting on how good of an idea this is, since Simon covered that in the link...
