step- limited pcr after tagmentation with Nextera XT... how many cycles?
I am using Nextera XT kit for library proepartion.
I`m following Illumina recommendations performing 12 cycles of step limited pcr after tagmentation.
However, I do not see a clear smear on agarose gel.
If I perform 13 cycles the smear is clearly visible.
I`ll try to go to the Bioanalyzer but....
has anybody experienced the same?
if I add one cycle to my pcr, can this spoil my data?
