    storing TruSeq libraries > 1 week

    Hello all,

    I recently had to travel unexpectedly and have had to keep some TruSeq stranded libraries in the -20 for more than the 1 week called for in the Illumina protocol. They're at 2 weeks now but have not yet been diluted to 10nm. I have more libraries to finish this week and hope to send them for sequencing by week's end. I'm inclined to believe that undiluted libraries are still OK for a few weeks but thought I'd ask for good measure. Thanks in advance!

    They should be fine. The libraries in double strand form are like any other DNA and store well at -20C. I have sequenced libraries stored for several months. Issues can raise when low concentrated normalised libraries are stored and clustered without re-quantification.

      Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
