    poly(A) and total RNA from the same sample?

    Dear everybody,

    I am looking for RNASeq data, ideally from a human cell culture, less ideally from a primary tissue, with the requirement that the data should be from a poly(A) selection but also from the total RNA of the very same input RNA. So basically a pair of data sets generated from the same sample. The platform isn't really important but the read length should be >50 bp.

    Is anybody aware of such datasets?

    Hi Narty,

    Did you have a look at the ABRF Study (here)? It does not contain total RNA, but depleted and degraded samples of the same cell cultures (UHRR).

        Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
