    which method for Ribo-seq liberary construction is better?

    Hi, all
    I saw some methods for liberary construction of Ribo-seq, some recommend to use small RNA liberary construction that need to ligate RPF with 3' and 5' adapter, and others also suggest to use circularization after 3' adapter ligation instead of 5' adapter ligation. So what's the difference between them? and which is better? is there any problem for final analysis results?

    I recommend circularization ... I found that if your input RPF is not enough, 3' and 5' adapters can be ligated directly ...
