How to set "CISTEMATIC_ROOT" of CISTEMATIC

Hi everyone:
I am new to CISTEMATIC as well as python. Could you please tell me how to set CISTEMATIC_ROOT in detail. Thanks
By the way, Could you please tell me how to add Cistematic to PYTHONPATH . Currently I just put the whole dir into a existed PYTHONPATH ("/bin/python2.6")
