Does anyone know if smear analysis will ever give a 100% area measurement? I'm trying to figure out my standard error upon detecting two regions; first region starting right after the first marker to 400 bp, second region 400 bp to right before the last marker. I'm measuring A-tailed DNA fragments compared to non A-tailed DNA at increasing concentrations of enzyme, hence increasing blunt-end ligated material. I split the regions in two portions, but they both don't add up to 100%, or will come close at 97-99%.
