  • Simultaneous DNA + RNA Extraction from Cells

    Does anyone have any knowledge or experience on the possibility of extracting DNA and RNA from the same cell sample(s)?

    I would like to isolate both the DNA and the RNA from the same cell(s) separately so that I can do both RNA and DNA sample preps. Not sure if there is a commercial kit out there for this or if this is even a possibility?

    Obligatory obvious answer; just split it in two and separate RNA and DNA from each half separately.

    Not very helpful if the sample is limiting though!

      You can extract DNA, RNA and protein from the same sample if you want. You can do that with Trizol or more commercial kits like Qiagen's Allprep. The Qiagen kits are a lot faster, but more expensive.

        Yes, for simultaneous extraction of DNA/RNA/small RNA you could for example use Qiagen Allprep DNA/RNA/microRNA kit. The protocol is fairly straight forward and the output quality is very good.

        The AllPrep DNA/RNA/miRNA Universal Kit is designed for simultaneous purification of DNA and total RNA, including miRNA, from cells and tissues

          Thanks very much, the Qiagen kit looks promising! I have another difficult question now!

          Does anyone know if it's possible to extract RNA + Chromatin (not just DNA) from the same cell population?

          I want to be able to do RNA-seq and CHIP-seq on the same population without having to split them and extract RNA from half and Chromatin from the other half.
