  • Axygen Mag PCR Clean-up VS. FragmentSelect beads

    Hi All,

    Was wondering if anyone has observed an increase in low fragment removal efficiency by switching from Axygen Mag PCR Clean-up beads to the FragmentSelect-I beads?

    From my reading, the Clean-up beads don't claim to remove anything above 125bp, but the FragmentSelect-I is essentially programmable.

    Bead clean-up products all work using the same principal. For more info I would suggest looking at this blog and comments:http://core-genomics.blogspot.com.au...eads-work.html

    One can utilize any of them for removing fragments in desired cut off or range. Some products works exactly at the same ratios as AMPure beads. For any specific application fragmented DNA can be used to find desired ratio of beads to sample for cut off. Fragmented DNA works better than DNA ladder as its distribution is continuous as would be in a randomly fragmented sample used for library prep.

      Thats good information about ladder vs. fragmented DNA, it was something I was thinking about looking up.

      I was of a similar opinion that many of the clean-up beads seem to share the same properties, but as both the Axygen Mag PCR Clean-up beads and the FragmentSelect-I beads are both from the same company (Axygen), perhaps there was an advantage of one over the other for tailoring size selection.

      Would you think one product offers advantages over the other from the same company?

        Hi Squall
        That is a new trend among this type of bead products manufacturers. For instance, Beckman in addition to AMPure XP for PCR clean-up also markets SPRIselect beads for size selection. I never had any issues using AMPure XP for double size selection. The claim is that size selection products are tested more vigorously to ensure batch to batch consistency. This is something that I would think that any good manufacturer should do without asking for a premium price. The manufacturer would be the best to be asked about advantages of one over other.
