  • Removal of carry-over EtOH after bead purification of DNA

    I recently purified some DNA (double-digest of genomic DNA) with SPRI beads, and I don't believe I let them dry long enough to get rid of excess EtOH used during the rinse step before eluting the DNA with HPLC water.

    I read somewhere that placing the DNA solution on a SpeedVac for ~ 2 min should get rid of the ethanol, but I'm skeptical. Would that actually work, especially with such dilute EtOH?

    For what it's worth, I just use a pipette, and even with a drying time of less than one minute, I haven't had any inhibition of my PCRs using Phusion as an enzyme, at least. Occasionally I'll see some weirdness on my Bioanalyzer trace, but it's hard to say whether that's the product of EtOH contamination.
