    NCBI FTP Server?

Do anyone here have trouble accessing NCBI FTP server for VCF files? The below link given in their site does not work.

    ftp://ftp.ncbi.nih.gov/snp/organisms/human_9606/VCF

I need hg19.vcf or 00-All.vcf.gz which was supposed to be at:

    ftp://ftp.ncbi.nlm.nih.gov/snp/organ.../00-All.vcf.gz

Can anyone let me know where to get this file?

    Thanks

Maybe their servers are down. The full command didn't work for me, but what you can do is go type:

    ftp ftp.ncbi.nih.gov

, And then just follow the path to the directory you specified by:

    cd <dir_name>

    Then use:

    > get <file_name>.gz

My internet connection is bad right now, so it's taking a while, but maybe yours is better and so it might work.

I wrote to them and it looks like I am to reach from the MAC Finders 'Connect to Server' but not through any browser. I thank you for trying.

Maybe it was a temporary problem? The .gz file is there, and can be downloaded from a browser (tried Firefox).

          Hi,

It is working now, I believe. I could get to the directory in firefox and read the readme file.

Nine Things a Sample Prep Scientist Thinks About Before Sequencing
              by SEQadmin2

I’m not a sequencing expert. I’m a purification scientist who uses NGS to evaluate workflows my group develops. With this perspective, we think about the sample first and the NGS workflow second. The sequencer is an exceptionally honest reporter, but it can only report on what you give it, so whether you get clean, interpretable data from an NGS workflow is largely determined before you begin.

Here are nine questions we think about, in roughly the order they matter, before...

From Collection to Sequencing: Why Sample Preparation and Preservation Define Sequencing Data
              by SEQadmin2

Data variability is still an issue in sequencing technologies despite the advances in reproducibility and accuracy of these platforms. But the problem does not originate in the sequencing itself, but in the previous steps, before the sample reaches the sequencer.

The first step is collection, followed by preservation and sample preparation for analysis. Most scientists overlook those steps, but not being careful might just be skewing the experiment’s results.
