    idba_ud Mink and Maxk

I am wondering how to set the mink and maxk in idba-ud. I have already set the read length to be 328 by editing kMaxShortSequence in src/sequence/short_sequence.h but the mink and maxk remains <124. How do i change the maxk to be greater than >124 cos I have long 2x300 reads and want a higher maxk value. just setting it on command line gives error i.e --kmax 250 gives error

    Thank you
    Isaac

Hey All, I didnt look properly enough before posting. I did find the answer to my question. Yu Peng gave this " You can modify constant kNumUint64 in src/basic/kmer.h to a bigger value (e.g. 6 or 8). This constant controls the number of 64 bit integer for holding k-mer."
    That answers my question.
    Thank you
    Isaac

Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.

Nine Things a Sample Prep Scientist Thinks About Before Sequencing
      by SEQadmin2

I’m not a sequencing expert. I’m a purification scientist who uses NGS to evaluate workflows my group develops. With this perspective, we think about the sample first and the NGS workflow second. The sequencer is an exceptionally honest reporter, but it can only report on what you give it, so whether you get clean, interpretable data from an NGS workflow is largely determined before you begin.

Here are nine questions we think about, in roughly the order they matter, before...
