  • Questions on Agilent SureSelect Indexing Kit

    Hi,

    does anybody have details on the new SureSelect Indexing Kits?
    I am interested in targeting 12 samples and pool and sequence them on 1 lane of Illumina GAII.

    My question is:

    - What is the maximum amount of DNA that I can target using a 55k baits SureSelect library? For one sample with a Paired-End approach one can target up to 6.6Mb. Do I have to divide this amount by the number of samples that I want to target (i.e. 6.6/12=0.55Mb max per sample)?

    IMHO, indexing do not influence the amount of DNA you can capture with one SureSelect library. There is still one library for the enrichment of one DNA sample. Usage of indexing adapters and primers only allows you to sequence up to 12 enriched samples on a single GAII lane. Unfortunately it do not allow you to enrich 12 samples using a single SureSelect library...
