    Kapa HiFi Polymerase strand displacement

    Hi all,

    does anyone know about literature evidence or product sheets that provide information on the strand displacement activity of the Kapa HiFi polymerase?

    I am working on optimising my assay and I am looking for a processive polymerase with very little strand displacement ability. I tried to look it up in datasheets, protocols, etc. but have not been able to find concrete information on that. It would be great if someone knows more about that...

    Generally speaking, high processivity is related to high strand displacement activity, so you won't find a polymerase that fits your requirements. From what I recall, the KAPA enzyme is an evolved Pfx/KOD enzyme and has very strong strand displacement activity and processivity.

    There was a review in TiBt that summarized the biochemical properties of various polymerases:



    But I personally think that the NEB site is pretty good:

      Kapa does not have strand displacement activity

      This is a little late for the above discussion, but just in case others search and find this thread... according to Kapa Tech Support, it does NOT have any strand displacement activity.

        I can confirm the message above, KAPA tech support was very clear about this.

            Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
