    Probe designing tips for targeted NGS

    Hello. I'm trying to analyse DNA methylation patterns of selected DMRs by the HiSeq system.
    I'm planning to use biotin labeled DNA probes to pull down target regions but not sure how to design them.
    Any advice and tips like length, GC ratio, # of biotin and etc. will be appreciated!

    I would suggest looking at following commercial products:

    1- SeqCap Epi developer enrichment kit (Roche)
    2- SureSelect Methyl-Seq kits (Agilent)

    #1 interrogates methylation status of both strands and captures bisulphite converted DNA
    #2 interrogates methylation status of one strand by capturing targets followed by conversion
