  • Clustering 1kb - 1.5kb fragments

    Hi Seq-sters!

    Curious whether anyone has tried this, heard of someone who has, or is interested in attempting? (Illumina platform)

    Any thoughts on:
    1. cBOT protocol manipulations?
    2. Fold increase in input concentration (assuming v3 chem)?

    Disclaimer: We are not interested in MP data, but if it works it could certainly be used for that application (depending on max cluster density, and coverage requirements).

    Never tried 1 Kb but 800-bp library worked fine without any special manipulations. The only modification that I did was the decrease of the input concentration (3 - 4.5 pM instead of 9-10; generally, I believe that for long libraries you should decrease the input concentration rather than increase). We got about 120 millions of reads on 2 lanes.
    And I'm interested very much in sequencing longer libraries. Please let us know if you try this!
