    Front manifold leaked

    Hi All:
I'm running a small sequencing center with 4 GAIIx; for the last few months we had experienced a high number of front manifold leaking, which resulted in a number of lost runs as well as down time. Illumina kept insisted that this is not a well known issue, but I don't believe that; therefore I would like to see if any one has the same problem and kindly let me know.
    thank you

We're yet to have any leaks in the actual GAIIx.

We did have leaks on our cluster station manifolds for many months before they replaced parts of it.

We have also have a leaking front manifold on our GaIIx. The Illumina technician attributed the problem to corrosion and we will get it replaced. We were instructed not to leave the flowcell in place between runs in the future as that could contribute to the problem [During our initial training we were instructed that we should leave the flowcell in place between runs].

The first two runs after the upgrade to GaIIx went well. In the third run, we did not see any problems when installing the flowcell, but we saw some bubbles in one of the lanes when looking at the images afterwards. Before the fourth run, we ran some tests and since we could not get the manifold to stop leaking we called for the Illumina technician.

Thank you Scott and Anders for quick reply. I'm still working with Illumina to correct the problem, but so far it's seem like we got a bad lot of front GA manifolds.

16S/ITS Profiling vs. Shotgun Metagenomics: What Changes in Your Workflow?
          by SEQadmin2

NGS has revolutionized clinical microbiology by offering new ways to identify and surveil pathogens without prior knowledge of the target. One popular approach is 16S rRNA and ITS amplicon sequencing, which targets specific microbial markers. But studies are increasingly incorporating shotgun metagenomics, a technique that sequences all DNA from a sample without targeting specific microbial markers. This approach allows more detailed taxonomic classification, detection...

Multiomic Liquid Biopsy: What DNA/RNA Panels Reveal That DNA Alone Can’t
          by SEQadmin2

Tumors contain complex mixtures of cell populations that evolve in response to treatment and host-specific factors. Tissue biopsies provide only static snapshots of a tumor’s state. But what if we could follow that evolution over time using a minimally invasive technique?

Liquid biopsies can detect tumor-derived material circulating in the bloodstream, providing a way to study treatment response, detect emerging resistance, and discover new biomarkers. Circulating
