  • Illumina Lab control

For people who had experience with illumina GA, what are the temperature, humidity, vibration and dust control like in your illumina room? Is it strict or its just like a normal wetlab setup? Do you think these factors can influence the sequencing results? Thanks alot in advance.

As any high precision system all those external factors can certainly affect the sequencing results. A minimal decent deployment environment will suffice to warranty those don't affect the process.

You'll have other things to worry about though.

We had severe problems with heat issues in our lab in the early days when we had a GA (GAI or GA 'classic'). Our lab was maintained at 23 degrees and it was too hot. We keep a portable air-conditioner blowing on it these days, although I think with the GAIIx we could probably do without it (but it doesn't do any harm, so we'll keep it for now!).

My instrument lab has had a picture of Alaska stuck on the door, I am not taking the hint from the people that have to work in there though! Does anyone know of a supplier of fleece lined labcoats?

Not to open new thread so writing in this one

In the preparation site PDF is pointed that Illumina recommends placing the GAIIx on mobile bench. So does GAIIx needs special anti-vibration table/bench?

@James what is the normal temp you set at the time of the run in the room

            Hi Vebaev,

You don't need an anti-vibration table. They recommend a movable bench just in case the service engineers need to access the back of the instrument. You don't really need it if it's accessible from all sides.

FWIW: We run our GAIIx in a room that fluctuates between 18 and 22 degrees C. We used to run an air-con unit on it to keep it cool, on the recommendation of Illumina, but we don't do it anymore, and it's fine.

            Cheers,

            Scott.

              Strategies for Sequencing Challenging Samples
              by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
