Miseq custom primer design, primer linker
Many papers have used a 2bp primer linker in 5’ end of the amplicon primers. As, I understand this is to prevent taxon specific PCR bias due to matches to the overhanger and/ or barcodes. I am wondering why 16S protocol did not add a linker.
