HELP!!!! How stable are Nextera Index primers?

The problem has been solved, thanks a lot!

They are probably fine, but if you don't want to take a chance, you could phone Illumina tech support and ask them. They might be willing to replace the primers.

I would simply test a few primers with index-PCR, and run on BA side by side.

they are quite robust, I have been freezing and thawing them probably 10 times or more and they were still fine. Unless you are using 50 ng of input DNA for your Nextera library prep, you have a huge excess of index primers anyway, so if part of them went bad, you are still safe.
We use the Nextera XT kit, start from <500 pg for tagmentation and dilute the primers 1:5 to ame them last longer. And, even then, if we don´t do a bead clean up afterwards, we still see a huge peak of unused adaptors in the prep.

16S/ITS Profiling vs. Shotgun Metagenomics: What Changes in Your Workflow?
          by SEQadmin2

NGS has revolutionized clinical microbiology by offering new ways to identify and surveil pathogens without prior knowledge of the target. One popular approach is 16S rRNA and ITS amplicon sequencing, which targets specific microbial markers. But studies are increasingly incorporating shotgun metagenomics, a technique that sequences all DNA from a sample without targeting specific microbial markers. This approach allows more detailed taxonomic classification, detection...

Multiomic Liquid Biopsy: What DNA/RNA Panels Reveal That DNA Alone Can’t
          by SEQadmin2

Tumors contain complex mixtures of cell populations that evolve in response to treatment and host-specific factors. Tissue biopsies provide only static snapshots of a tumor’s state. But what if we could follow that evolution over time using a minimally invasive technique?

Liquid biopsies can detect tumor-derived material circulating in the bloodstream, providing a way to study treatment response, detect emerging resistance, and discover new biomarkers. Circulating
