Is anyone aware of NGS-based estimates of mutation rates for mammalian cell culture? I know that NGS was used to estimate germline mutation rate in humans but I am interested in the mutation rate of human cells in culture. So this would be somatic/mitotic mutation rate.

expected mutation rates?

I have no idea on studies using NGS, but some independent OLD papers suggests in vitro cell culture increase mutation rate by 10x
in vivo retionblastoma: 10x-8 order
(http://www.ncbi.nlm.nih.gov/pmc/arti...00396-0056.pdf)
in vivo Y chromosome
in vitro germlien: 10x-8, approaching 10-7
(http://www.pnas.org/content/89/15/7036.full.pdf)
Normal vs Malignant lymphocyte, 10x in malignant

Somatic mutation: 10-7

There is a recent nextgen study that puts human y-chromosome mutation rate at 10^-8 but I too have not found a NGS-derived cell culture mutation rate. So I suppose this does put cell culture at 10X the mutation rate of the human germline.Originally posted by marcowanger View PostI have no idea on studies using NGS, but some independent OLD papers suggests in vitro cell culture increase mutation rate by 10x

Yes, I also found that paper, I remember it's in CELL press / Curr Bio. Anyway, I could not find any NGS study on somatic vs germline mutation rate too.Originally posted by genericforms View PostThere is a recent nextgen study that puts human y-chromosome mutation rate at 10^-8 but I too have not found a NGS-derived cell culture mutation rate. The immune system’s power comes from its genetic diversity, allowing myriad threats to be neutralized through first recognizing foreign antigens. That diversity is also what makes the immune system so difficult to study. Recent advances in sequencing technology and computational biology, however, are giving researchers new tools to understand immune responses and immune-related diseases in greater detail.
This convergence of genetics, immunology, and computation...-
